Seems strange to me that they would ask you to do a demo lesson for an assistant position. That being said, numbers seem sort of advanced when you have 2 year olds in the group and you could be setting yourself up for failure. Could you maybe focus on the color learning? Perhaps have the kids sort pompoms by their color and drop them into the corresponding color jar or tube (you could roll colored paper). That would have two objectives: sort objects by one attribute (color) and develop fine motor coordination. Motor skills just seem more relevant at that young age than number sense.

Your plan seems overly complicated. As an assistant you
Would be followwing the teacher’s plan. Design aomething... counting, phonemic base that could link to most lessons connections

They asked you to prepare a demo lesson for this? Because I used to work in daycare and by "2nd interview", it was just to see how you fit in in the classroom and to see how you were with the kids. They didn't even watch you. They ask the teacher in the room how you did. You pretty much followed the teacher's lead and did whatever she did. If it was free play time, you just played with them.
